ABSTRACT

Abstract Objective To evaluate the systemic effect of Hancornia speciosa latex on bone neoformation and mineralization in rats. Methods For that, the latex was first collected, and its composition was analyzed. A total of 30 male Wistar rats were used, which were simultaneously submitted to two surgical procedures: extraction of an incisor and creation of a defect with 2 mm in diameter in the parietal bone. The rats were divided into two groups: systemic control (SC) systemic latex (SX) which were administered, orally and daily, 1.5 mL of water or a solution containing 50% of water and 50% of latex by gavage, respectively. After 15 days of the treatment, the animals were euthanized and their samples were collected. Results The results were statistically analyzed, and the level of significance was set at 0.05. We showed that H. speciosa latex contained calcium. The oral and daily administration of the latex for 15 days increased the contents of calcium and phosphorus in the basal bone and newly-formed bone in the mandibular alveolus of rats. Conclusion The present was a pioneer study demonstrating the potential of H. speciosa latex in increasing bone mineralization. Our results may aid in the conception and development of a natural drug.


Resumo Objetivo Avaliar o efeito sistêmico do látex de Hancornia especiosa na neoformação óssea e mineralização em ratos. Métodos Para isso, primeiro o látex foi coletado, e sua composição foi analisada. No estudo, foram utilizados 30 ratos Wistar machos submetidos simultaneamente a dois procedimentos cirúrgicos: extração de incisivo e criação de um defeito de 2 mm de diâmetro no osso parietal. Os ratos foram divididos em dois grupos: controle sistêmico (CS) e látex sistêmico (XS), aos quais foi administrado, oral e diariamente, 1,5 mL de água ou uma solução contendo 50% de água e 50% de látex por gavagem, respectivamente. Após 15 dias do tratamento, os animais foram eutanizados, e suas amostras, coletadas. Resultados Os resultados foram analisados estatisticamente, e o nível de significância foi fixado em 0,05. Mostramos que o látex de H. speciosa continha cálcio. A administração oral e diária deste látex por 15 dias aumentou o conteúdo de cálcio e fósforo de osso basal e de osso recém-formado no alvéolo mandibular de ratos. Conclusão Este foi um estudo pioneiro, que demonstrou o potencial do látex de H. speciosa no aumento da mineralização óssea. Nossos resultados podem ajudar na concepção e no desenvolvimento de uma droga natural.

ABSTRACT

@#Eleven alkaloids were isolated from the twigs and leaves of Ervatamia pandacaqui using chromatographic methods of silica gel, Sephadex LH-20, ODS, and HPLC.Their structures were elucidated by physical,chemical and spectroscopic methods and determined as voacristine 7-hydroxyindolenine (1),iboxygaine (2), 19S-hydroxyibogamine (3), 3-oxotabersonine (4), perivine (5), pericyclivine (6), rhazinalinol (7), geissoschizol (8), 3, 14-dihydroolivacine (9), vallesamine (10), and conolobine A (11), respectively.All compounds were isolated from this plant for the first time.

ABSTRACT

ABSTRACT: Desert rose (Adenium obesum) is a succulent plant that is native to the Arabian Peninsula. It has great commercial value with exuberant structure and flowering. It can be propagated by seeds to ensure maximum development of the caudex. Despite its high economic value, studies on its propagation are still required. The aim of this study was to evaluate seed germination performance and seedling development of desert rose seedlings in different substrates. Treatments comprised five different substrate compositions: S1 (Soil), S2 (Sand), S3 (Carolina Soil commercial substrate), S4 (Sand + Carolina Soil), and S5 (Soil+ Carolina Soil). The following were evaluated using uni- and multi-variate approaches: germination, first count, germination speed index, caudex diameter, number of leaves, seedling length, root length, shoot length, and fresh mass and dry mass. Sand is the ideal substrate for the germination of A. obesum seeds, while for forming seedlings, mixtures of the Carolina Soil commercial substrate with sand or soil should be used.


RESUMO: A rosa do deserto (Adenium obesum) é uma planta suculenta nativa da Península Arábica, apresenta grande valor comercial, com estrutura e floração exuberante. Sua propagação pode ser por meio de sementes para garantir o maior desenvolvimento do caudex. Apesar do alto valor econômico ainda se requerem estudos para sua propagação. O objetivo foi avaliar o desempenho germinativo de sementes e desenvolvimento de mudas de rosa-do-deserto em diferentes substratos. Os tratamentos constaram em cinco diferentes composições de substrato: S1 (Terra), S2 (Areia), S3 (substrato comercial Carolina Soil), S4 (Areia+ Carolina Soil) e S5 (Terra + Carolina Soil). Foram avaliados com abordagem uni e multivariada: germinação, primeira contagem, índice de velocidade de germinação, diâmetro do caudex, número de folhas, comprimento da plântula, comprimento da raiz, comprimento da parte aérea, massa fresca e massa seca. A areia é o substrato ideal para a germinação de sementes de A. obesum, enquanto que para a formação das mudas deve-se utilizar as misturas do substrato comercial Carolina Soil com areia ou terra.

ABSTRACT

Introduction: Rice-field rats are one of the most important pests because it can give large losses in all planting seasons including the storehouse. Synthetic rodenticide is the most commonly used of chemical technique for controlling rice-field rats. The application of these materials indirectly causes negative impacts; one of them is for the environment. As an alternative for controlling rice-field rats, natural materials can be used as a repellent. Objective: To examine the effects of methanol extract of Plumeriarubra leaves on metabolism, daily activity patterns, and its potency as a repellent of the rice-field rat. Methods: The experiments were conducted at the Laboratory of Pests, UniversitasPadjadjaran involves choice test (T-maze arena), and the Laboratory of Rats, Indonesian Center for Rice Research involves no-choice test (metabolic cage) from February until May 2019. The observations including food (g), water consumption (ml), feces production (g), urine production (ml), body weight (g), and its changes (%), also the daily activities (time spent for locomotion, foraging, and resting).The treatment was done with three replications for twelve mature male and twelve mature non-pregnant females. Data experiments analysis followed by a T-test. Results: Rice-field rats on the T-Maze arena avoided consuming food and beverage that close to methanol extract of Plumeriarubra leaves treatment. The treatment of methanol extract of Plumeria leaves in metabolic cage caused metabolic disorder of rice-field rat, which was significantly indicated by the decrease of the average consumption of food by 2.28 g and excretion of feces by 0.34 g, and also the increase of average consumption of beverage by 3.89 ml, excretion of urine by 3.15 ml, and body weight by 6.67 g. The treatment also caused daily activity patterns disorder of rice-field rats, which was significantly indicated by the increase of the average percentage of time for movement activities (locomotion) by 7.64 % and the decrease of time for eating and drinking activities (foraging) by 16.46 %. Conclusion: Methanol extract of Plumeria leaves affects a repellent for the rice-field rat.


Introducción: Las ratas arroceras son una de las plagas más importantes porque pueden producir grandes pérdidas en todas las temporadas de siembra, incluso en el almacenaje. La técnica química más utilizada para controlar las ratas de los arrozales es el raticida sintético. Sin embargo, la aplicación de estos químicos provoca indirectamente impactos negativos, por ejemplo, en el ambiente. Una alternativa para controlar la rata arrocera es la utilización de compuestos naturales como repelentes. Objetivo: Examinar los efectos del extracto metanólico de hojas de Plumeria rubra sobre el metabolismo, los patrones de actividad diaria en las ratas arroceras y su potencial como repelente. Métodos: Los experimentos se llevaron a cabo en Laboratory of Pests, UniversitasPadjadjaran usando la prueba T-maze arena, y en Laboratory of Rats, Indonesian Center for Rice Research usando la prueba metaboliccage, desde febrero hasta mayo 2019. Las observaciones incluyeron consumo de alimentos (g), consumo de agua (ml), producción de heces (g), producción de orina (ml), peso corporal (g) y cambios (%), además actividades diarias (tiempo dedicado a la locomoción, búsqueda de alimento, y reposo). El tratamiento se realizó con tres repeticiones para 12 machos maduros y 12 hembras maduras no gestantes. Los análisis de experimentos de datos se realizaron con la prueba T. Resultados: Las ratas arroceras en la T-maze arena evitaron consumir alimentos y bebidas cercanos al extracto de metanol de hojas de Plumeria rubra. El tratamiento del extracto metanólico de hojas de Plumeria rubra en la prueba metaboliccage provocó un trastorno metabólico en estas ratas, lo cual se demostró significativamente en la disminución del consumo promedio de alimento en 2.28 g y la excreción de heces en 0.34 g, además en el aumento del consumo promedio de bebida en 3.89 ml, excreción de orina en 3.15 ml y peso corporal en 6.67 g. El tratamiento también provocó un trastorno en los patrones de actividad diaria de las ratas, lo cual fue demostrado por el aumento significativo en el porcentaje promedio de tiempo para actividades de movimiento (locomoción) en un 7.64 % y la disminución del tiempo para comer y beber (búsqueda de alimento) en un 16.46 %. Conclusión: El extracto metanólico de hojas de Plumeria rubra tiene un efecto repelente en las ratas arroceras.

ABSTRACT

Four alkaloids were isolated from the total alkaloids of the twigs and leaves of Alstonia yunnanensis (Apocynaceae) by using silica gel, ODS, Sephadex LH-20, and HPLC chromatography. Structures were determined by physical, chemical and spectroscopic methods and identified as N4-methylpseudoakuammigine (1), pseudoakuammigine (2), vinorine (3), picraline (4). Among them, compound 1 is a new monoterpenoid indole alkaloid.

ABSTRACT

To ensure the safety of medications, it is vital to accurately authenticate species of the Apocynaceae family, which is rich in poisonous medicinal plants. We identified Apocynaceae species by using nuclear internal transcribed spacer 2 (ITS2) and psbA-trnH based on experimental data. The identification ability of ITS2 and psbA-trnH was assessed using specific genetic divergence, BLAST1, and neighbor-joining trees. For DNA barcoding, ITS2 and psbA-trnH regions of 122 plant samples of 31 species from 19 genera in the Apocynaceae family were amplified. The PCR amplification for ITS2 and psbA-trnH sequences was 100%. The sequencing success rates for ITS2 and psbA-trnH sequences were 81% and 61%, respectively. Additional data involved 53 sequences of the ITS2 region and 38 sequences of the psbA-trnH region were downloaded from GenBank. Moreover, the analysis showed that the inter-specific divergence of Apocynaceae species was greater than its intra-specific variations. The results indicated that, using the BLAST1 method, ITS2 showed a high identification efficiency of 97% and 100% of the samples at the species and genus levels, respectively, via BLAST1, and psbA-trnH successfully identified 95% and 100% of the samples at the species and genus levels, respectively. The barcode combination of ITS2/psbA-trnH successfully identified 98% and 100% of samples at the species and genus levels, respectively. Subsequently, the neighbor joining tree method also showed that barcode ITS2 and psbA-trnH could distinguish among the species within the Apocynaceae family. ITS2 is a core barcode and psbA-trnH is a supplementary barcode for identifying species in the Apocynaceae family. These results will help to improve DNA barcoding reference databases for herbal drugs and other herbal raw materials.

Abstract Objective The present work aimed to evaluate the systemic effect of H. speciosa latex on bone neoformation. Methods For this, the latex was collected and diluted to 3% and 50%. A total of 28 Wistar rats were submitted to surgery to create a 5 mm diameter defect in the parietal bone. This experiment was conducted in 2 different periods: 1 and 2. For each period, the rats were divided into 3 groups: Control Group, Latex3 Group, and Latex50 Group, which received, respectively, daily administrations of 0.5 mL of distilled water, latex to 3% and latex to 50% by gavage, orally. The rats of periods 1 and 2 were euthanized, respectively, 15 and 30 days after the surgery, and the calvaria was collected. The results were analyzed using the ANOVA and Tukey tests; the significance level was 0.05. Results We show that, in each analyzed period, the experimental groups had the same amount of newly formed bone in the calvaria defect. Conclusion We conclude that daily and oral administrations of H. speciosa latex to 3% and to 50% over a period of 15 and 30 days does not contribute to the increase of the area of the newly formed bone in the calvaria defect.


Resumo Objetivo Este trabalho objetivou avaliar o efeito sistêmico do látex de H. speciosas obre a neoformação óssea. Métodos Para isso, o látex foi coletado e diluído a 3% e a 50%. Um total de 28 ratos Wistar foi submetido a cirurgia para a criação de um defeito de 5 mm de diâmetro no osso parietal. Esse experimento foi conduzido em dois períodos distintos: 1 e 2. Para cada período, os ratos foram divididos em 3 grupos: Grupo Controle, Grupo Látex3 e Grupo Látex50 que receberam, respectivamente, administrações diárias de 0,5 mL de água destilada, látex a 3% e látex a 50% por gavagem, via oral. Os ratos dos períodos 1 e 2 foram eutanasiados, respectivamente, 15 e 30 dias após a cirurgia e a calvária foi coletada. Os resultados foram analisados utilizando os testes ANOVA e Tukey; o nível de significância estabelecido foi 0,05. Resultados Mostramos que, em cada período analisado, os grupos experimentais tiveram a mesma quantidade de osso neoformado no defeito da calvária. Conclusão Portanto, concluímos que administrações diárias e orais do látex de H. speciosa a 3% e a 50% durante um período de 15 e 30 dias não contribui para o aumento da área do osso neoformado no defeito da calvária.

Objective: The objective of the present research work had been done to evaluate the toxicity of crude extract of Carissa spinarum in Swiss albino mice. Methods: In studying the toxicity, the Organization for Economic Cooperation and Development (OECD) guidelines were used. Experimental animals (mice), five mice in each, were grouped into four groups; three experimental groups and one negative control. In studying the acute toxicity, 2000, 3000 up to 5000 mg/kg crude plant extract was given orally using standard intragastric oral gavages. For acute toxicity, a single dose was given and gross behavioral changes were recorded. In sub-acute oral toxicity test, Carissa spinarum crude extract was given to the mice by standard intragastric oral gavages at doses of 500, 750 and 1000 mg/kg body weight of hydro-methanolic extract and 200, 600 and 1000 mg/kg of body weight of chloroform extract in every single to 28 d and various hematological and physical parameters were recorded. Results: In acute toxicity, the given dose of the plant extract did not produce significant physical and behavior changes up to the dose of 5000 mg/kg extracts. In addition, no death was occurred in the given doses. In sub-acute toxicity studies of the hydro-methanolic and chloroform extracts, there was no recorded significant change (p>0.05) of hematological and physical parameters in the treated groups when compared to the control groups. Conclusion: from the present study it was revealed that the crude extract of the plant did not produce any significant toxicological effect in the experimental animals and this supports the use of the plant in folk medicines.

Objective: To study the chemical constituents in the twigs and leaves of Ervatamia officinalis. Methods: The chemical constituents were isolated from the total alkaloids of the twigs and leaves of Ervatamia officinalis by using silica gel, ODS, Sephadex LH-20, and HPLC. The chemical structures of the compounds were identified based on their physical and chemical properties and spectroscopic methods. Results: Fourteen alkaloids were isolated and identified as 7S-coronaridine hydroxyindolenine (1), 7S-voacangine hydroxyindolenine (2), coronaridine (3), 19S-heyneanine (4), voacangine (5), tabernaemontanine (6), dregamine (7), 3-(2-oxopropyl) coronaridine (8), 3-oxovoacangine (9), voastrictine (10), 16R,19E-isositsirikine (11), 16R,19Z-isositsirikine (12), 16R,19E-isositsirikine N4-oxide (13), and geissoschizol (14). Conclusion: Compounds 8-14 are obtained from this plant for the first time.

ABSTRACT

Four new corynanthe-type alkaloids, meloslines C-F (1-4), together with four known ones (5-8) were isolated from the roots of Alstonia scholaris. Their structures including absolute configurations were elucidated by extensive spectroscopic analysis and electronic circular dichroism (ECD) calculation. Compounds 1 and 2 exhibited potent vasorelaxant activity on endothelium-intact renal arteries precontracted with KCl.

ABSTRACT In the past decades, desert rose has become a very popular ornamental plant, especially among collectors, due to its exotic and sculptural forms. However, it has been grown on a commercial scale only recently, and little is known about how to best manage it as a container-grown plant, or even which potting medium (substrate) to recommend. Therefore, the aim of this study was to investigate the interactions between potting media and irrigation levels for growing desert rose as a potted ornamental plant. The experiment was conducted in a greenhouse using a 6 x 2 factorial arrangement with six replications, six potting media and two irrigation levels. The mixes were characterized by measuring their physical properties, specifically the density and water retention capacity (WRC), as well as chemical properties, such as the pH and electrical conductivity (EC). After 210 days, plant growth and plant water consumption were evaluated and measured. A lower dry density for the vermiculite mixes was observed in comparison to that for the sand mixes. However, WRC ranged from 428 to 528 mL L-1 among the mixes, values considered close to ideal. In general, plant growth exhibited higher increases in mixes consisting of coconut fiber + sand or vermiculite, regardless of the irrigation level. Mixes of vermiculite + coconut fiber and sand + coconut fiber can be used to grow desert rose in pots, as long as irrigation is used to maintain the moisture content of the potting medium (mix) between 60-70% and 80-90% of the WRC.


RESUMO Na última década a rosa do deserto ganhou destaque no mercado de flores, principalmente entre colecionadores, devido às suas formas exóticas e esculturais; porém, o cultivo da espécie em escala comercial é bastante recente e pouco se sabe acerca do manejo da cultura, inclusive no que diz respeito à recomendação de substratos. Assim, objetiva-se nesse trabalho estudar a interação entre substratos e níveis de irrigação no desenvolvimento de rosa do deserto em vaso. Conduziu-se o experimento em casa de vegetação em esquema fatorial 6 x 2, com seis repetições, adotando-se seis substratos e dois níveis de irrigação. As misturas foram caracterizadas mensurando-se suas propriedades físicas (densidade e capacidade de retenção de água - CRA) e químicas (pH e condutividade elétrica - CE). Aos 210 dias de cultivo avaliou-se o crescimento das plantas; além do consumo de água pela cultura. Para as misturas compostas por vermiculita verificou-se menor densidade seca em relação às compostas por areia; no entanto, a CRA variou de 428 a 528 mL L-1 entre as misturas, valores próximos aos considerados ideais. De modo geral, o crescimento das plantas teve maiores incrementos nas misturas compostas por fibra de coco mais areia ou vermiculita, independente do nível de irrigação. As misturas vermiculita + fibra de coco e areia + fibra de coco podem ser empregadas para o cultivo em vaso de rosa do deserto, quando se mantiver os níveis de irrigação entre 60-70% e 80-90% da CRA das misturas.

To inverstigate the alkaloids from the twigs and leaves of Ervatamia pandacaqui, eleven known alkaloids were isolated by silica gel, Sephadex LH-20, and ODS column chromatography, as well as RP-HPLC. Their structures were elucidated by UV, IR, MS, and NMR spectral data as coronaridine (1), 3-oxocoronaridine (2), 19S-heyneanine (3), 19R-heyneanine (4), voacangine (5), 3-oxovoacangine (6), voacristine (7), 19-epi-voacristine (8), iso-voacangine (9), coronaridine 7-hydroxyindolenine (10), and voacangine 7-hydroxyindolenine (11). Compounds 1-11 were isolated from E. pandacaqui for the first time.

OBJECTIVE: To study the chemical constituents of Catharanthus roseus. METHODS: Various column chromatograghic methods on silica gel, Rp-18, and Sephadex LH-20 were applied for the isolation and purification of the 95% ethanol extract. The structures were elucidated by their physicochemical properties and spectral data. RESULTS: Twenty-two compounds were obtained and identified as ursolic acid (1), daucesterol (2), tetrahydroalstonine (3), 7α-hydroxy-β-sitosterol (4), vindoline (5), β-sitosterol (6), aurantiamide acetate (7), lochnericine (8), oleanolic acid (9), ajmalicine (10), (22E, 24R)-ergosta-7, 22-dien-3β, 5α, 6β-triol (11), betulinic acid (12), stigmasterol (13), quercetin (14), keampferol (15), vindorosine (16), catharanthine (17), diaaurantiamide acetate (18), reserpine (19), panarine (20), serpentine (21), and 16R-E-isositsirikine (22). CONCLUSION: Compounds 4, 7, 11, 13, 18, and 20 are isolated from C. roseus for the first time.

ABSTRACT

Objective: To study the alkaloids from Ervatamia hainanensis. Methods: The alkaloids were isolated and purified by silica gel, MCI, Sephadex LH-20 column chromatography, and semi-preparative HPLC, and their structures were elucidated by physical and spectroscopic analysis. Results: Twelve alkaloids were obtained and identified as coronaridine (1), 19-epi-heyneanine (2), 9,10-dimethoxycoronaridine (3), vobasine (4), vobasine N(4)-oxide (5), 3-oxo-19-epi-heyneanine (6), strictamine (7), deacetylakuammiline (8), pandine (9), rhazicine (10), rhazicine N(4)-oxide (11), and rhazimine (12). Conclusion: Compounds 8 and 10-12 are isolated from the genus Ervatamia Stapf for the first time, while compounds 3 and 5-7 are firstly obtained from E. hainanensis.

Chemical investigation of the dichloromethane extracts of Hoya diversifolia Blume led to the isolation of β-amyrin cinnamate (1), squalene (2), β-sitosterol (3), a mixture of β-amyrin (4a), α-amyrin (4b) and lupeol (4c) in a 4:2:1 ratio and saturated hydrocarbons from the leaves; and 2, taraxerol (5), lupeol cinnamate (6), and a mixture of 3 and stigmasterol (7) in a 2:1 ratio from the stems. The structures of 1-7 were identified by comparison of their NMR data with those reported in the literature.

ABSTRACT

Hancornia speciosa Gomes is a fruit species belonging to the Apocynaceae family and holds social, cultural and economic potential mostly due to its use in composition of many food industry products and the consumption its fruits in natura. Several aspects regarding their propagation need further studies, since the species is undergoing a continuous process of domestication. The objective was to obtain an in vitro protocol for indirect organogenesis and rooting with subsequent acclimatization of H. speciosa plants. To obtain indirect organogenesis, internodal segments were inoculated in WPM culture medium gelled with 7 g L-1 agar, added with 30 g L-1 sucrose, 0.4 g L -1 PVP, and supplied with different concentrations of 2,4-D (0.0; 2.46; 7.38; 12.30; and 17.22 µM), BAP (0.0; 4.92; 9.84; 14.76; and 19.68 µM), and TDZ (0.0; 4.92; 9.84; 14.76; and 19.68 µM). For the in vitro rooting, shootings with approximately 6.0 cm diameter length kept for 15 days in WPM medium with no plant growth regulator and, afterwards, subjected to treatments with different auxins (Control; 9.84 µM IBA; 9.84 µM NAA; and 9.84 µM IAA) as well as the combination among them, in order to verify their effects on percentage of rooting (%), number of roots, and average length of the largest root (cm). The formation of calluses was observed in all explants subjected to the concentrations of the regulators tested. The highest shooting regeneration occurred with 7.38 µM 2,4-D (73%). The highest percentage of shoot rooting (80%) and roots with the largest length (1.3 cm) were found in the culture medium with the combination of 4.92 µM NAA and 4.92 µM IBA. The in vitro regeneration of H. speciosa is feasible. The acclimatization of rooted shoots in Trospstrato® was accomplished with successful and 100% survival of plant material was observed during this stage.


Hancornia speciosa Gomes é uma frutífera pertencente à família Apocynaceae e possui grande potencial social, cultural e econômico, principalmente devido à utilização de seus frutos na composição de diversos produtos do setor alimentício e para consumo in natura. Vários aspectos de sua propagação necessitam de maiores estudos, uma vez que a espécie passa por um contínuo processo de domesticação. Objetivou-se obter um protocolo de organogênese indireta e de enraizamento in vitro, com posterior aclimatização das plantas de H. speciosa. Para a obtenção de organogênese indireta, segmentos internodais foram inoculados em meio de cultivo WPM gelificado com ágar 7 g L-1, acrescido de 30 g L-1 de sacarose, 0,4 g L-1 de PVP e suplementado com diferentes concentrações de 2,4-D (0,0; 2,46; 7,38; 12,30 e 17,22 µM), BAP (0,0; 4,92; 9,84; 14,76 e 19,68 µM) e TDZ (0,0; 4,92; 9,84; 14,76 e 19,68 µM). Para o enraizamento in vitro, foram utilizadas brotações com aproximadamente 6,0 cm de comprimento, mantidas por 15 dias em meio WPM isento de fitorreguladores e, em seguida, submetidas a tratamentos com diferentes auxinas (Controle; 9,84 µM de AIB; 9,84 µM de ANA e 9,84 µM de AIA), bem como as combinações entre estes reguladores (4,92 µM AIA + 4,92 µM AIB; 4,92 µM ANA + 4,92 µM AIB; 4,92 µM ANA + 4,92 µM AIA). Foi observada formação de calos em todos os explantes submetidos às concentrações dos reguladores testados. A maior regeneração de brotação ocorreu com 7,38 µM de 2,4-D (73%). Maior porcentagem de enraizamento das brotações (80%) e raízes com maior comprimento (1,3 cm) foi verificada em meio de cultivo contendo a combinação de 4,92 µM de ANA e 4,92 µM de AIB. É possível a regeneração in vitro de H. speciosa. A aclimatização foi realizada com sucesso, com 100% de sobrevivência das plantas.

ABSTRACT

As plantas do gênero Marsdenia, família Apocynaceae (Asclepiadoideae), apresentam distribuição mundial, embora se encontre predominantemente nas regiões tropicais. Diversas espécies de Marsdenia são usadas na medicina tradicional asiática para o tratamento de dores reumáticas, inflamação, asma, sífilis e câncer. Vários estudos fitoquímicos mostraram que elas contêm glicosídeos esteroides, principalmente derivados de pregnano, mas também alcaloides esteroides, triterpenos e conduritoles. No entanto, casos espontâneos de intoxicação ocorreram em diversas espécies animais, e podem ocorrer em humanos. Assim, as plantas do gênero Marsdenia apresentam enorme potencial para descoberta de novos compostos para uso como medicamentos.

Chemical investigation of the dichloromethane extracts of Hoya buotii Kloppenb. afforded taraxerone (1), taraxerol (2), a mixture of β-sitosterol (3a) and stigmasterol (3b) in about 2:1 ratio, and a mixture of α-amyrin cinnamate (4a) and β-amyrin cinnamate (4b) in about 1:2 ratio from the stems; 1, 2, and 3a from the roots; a mixture of 4a and 4b in about 3:2 ratio from the flowers; and 3a, squalene (5) and saturated hydrocarbons from the leaves. The structures of 1-5 were identified by comparison of their NMR data with those reported in the literature.

ABSTRACT

Nerium oleander é uma planta ornamental responsável por intoxicações em animais e humanos. Todas as partes da planta contém glicosídeos cardiotóxicos, principalmente a oleandrina. Alguns autores apontam que a toxicidade da planta apresentaria variação em função da cor da flor. O objetivo deste trabalho foi determinar se existe variação na concentração de oleandrina nas folhas de N. oleander de exemplares da planta com diferentes cores de inflorescências. Foram coletadas 10 amostras de folhas para cada tipo de flor (branca, rosea e vermelha). Os níveis de oleandrina foram determinados por meio de HPLC-UV após extração com metanol, precipitação com acetato de chumbo e separação em cartucho de C18. A média da concentração obtida em todas as folhas analisadas foi de 4,89mg g-1, sendo 6,20±4,08mg g-1 na variedade de flores brancas, 4,16±3,44mg g-1 na de flores roseas e 4,31±1,99mg g-1 na de flores vermelhas. As concentrações de oleandrina não apresentaram diferença estatisticamente significante entre as variedades de flores.


Nerium oleander is an ornamental plant responsible for poisoning in animals and humans. All parts of the plant contain cardiotoxic glycosides, mainly oleandrin. Some authors suggest that the toxicity of the plant would present a dependence on flower color. The objective of this study was to determine whether there is variation in the concentration of oleandrin in the leaves of N. oleander specimens of different colors of flowers. Ten samples of leaves from each variety of flower (white, pink and red) were collected. Oleandrin levels were determined by HPLC-UV after extraction with methanol, precipitation with lead acetate and separation with C18 cartridge. The average concentration obtained in all analyzed leaves was 4.89mg g-1, being 6.20±4.08mg g-1 in variety of white flowers, 4.16±3.44mg g-1 in pink flowers, and 4.31±1.99mg g-1 in red flowers. The concentrations of oleandrin did not show statistically significant difference between the varieties of flowers.

ABSTRACT

Chemical investigation of the dichloromethane extracts of Hoya multiflora Blume led to the isolation of lupeol (1a), α-amyrin (1b), β-amyrin (1c), lupeol acetate (2a), α-amyrin acetate (2b), and β-amyrin acetate (2c) from the stems; and 1b, bauerenol (3), squalene (4), lutein (5), β-sitosterol (6a), and stigmasterol (6b)from the leaves. The structures of 1-6 were identified by comparison of their 1H and/or13C NMR data with those reported in the literature.
